One of\nthe immediate reactions to stress signals is a reduction of protein synthesis (translation).\nUntil now, it was difficult to measure such acute cellular changes. As reported\nin the latest online issue of the renowned journal <em>Molecular Cell<\/em>, researchers at Goethe University have now developed\na method overcoming this hurdle.<\/p>\n\n\n\n<p>The team led by biochemist Dr. Christian\nM\u00fcnch, who heads an Emmy Noether Group, employs a simple but extremely\neffective trick: when measuring all proteins in the mass spectrometer, a booster\nchannel is added to specifically enhance the signal of newly synthesised\nproteins to enable their measurement. Thus, acute changes in protein synthesis\ncan now be tracked by state-of-the-art quantitative mass spectrometry.<\/p>\n\n\n\n<p>The idea emerged because the team wanted\nto understand how specific stress signals influence protein synthesis. &#8222;Since\nthe amount of newly produced proteins within a brief time interval is rather\nsmall, the challenge was to record minute changes of very small percentages for\neach individual protein,&#8220; comments group leader M\u00fcnch. The newly developed\nanalysis method now provides his team with detailed insight into the molecular\nevents that ensure survival of stressed cells. The cellular response to stress\nplays an important role in the pathogenesis of many human diseases, including cancer\nand neurodegenerative disorders. An understanding of the underlying molecular\nprocesses opens the door for the development of new therapeutic strategies.<\/p>\n\n\n\n<p>&#8222;The method we developed enables highly\nprecise time-resolved measurements. We can now analyse acute cellular stress\nresponses, i.e, those taking place within minutes. In addition, our method\nrequires little material and is extremely cost-efficient,&#8220; M\u00fcnch explains.\n&#8222;This helps us to quantify thousands of proteins simultaneously in defined\ntime spans after a specific stress treatment.&#8220; Due to the small amount of material\nrequired, measurements can also be carried out in patient tissue samples, facilitating\ncollaborations with clinicians. At a conference on Proteostatis (EMBO) in\nPortugal, PhD student Kevin Klann was recently awarded with a FEBS journal poster\nprize for his presentation of the first data produced using the new method. The\nyoung molecular biologist demonstrated for the first time that two of the most\nimportant cellular signaling pathways, which are triggered by completely\ndifferent stress stimuli, ultimately results in the same effects on protein\nsynthesis. This discovery is a breakthrough in the field.<\/p>\n\n\n\n<p>The project is funded by the European Research\nCouncil (ERC) as part of Starting Grant &#8222;MitoUPR&#8220;, which was awarded\nto M\u00fcnch for studying quality control mechanisms for mitochondrial proteins. In\naddition, Christian M\u00fcnch has received funding within the German Research\nFoundation\u2019s (DFG, <em>Deutsche\nForschungsmeinschaft<\/em>) Emmy Noether Programme and is a member of the Johanna\nQuandt Young Academy at Goethe. Since December 2016, he has built up a group on\n&#8222;Protein Quality Control&#8220; at the Institute for Biochemistry II at Goethe\nUniversity\u2019s Medical Faculty, following his stay in one of the leading\nproteomic laboratories at Harvard University. <\/p>\n\n\n\n<p class=\"has-background has-very-light-gray-background-color\"><strong>Publication:<\/strong><br>Klann K, Tascher G, M\u00fcnch C.
