Even more detailed insights into the cell will be possible in future with the help of a new development in which Goethe University was involved: Together with scientists from Israel, the research group led by Professor Harald Schwalbe has succeeded in accelerating a hundred thousand-fold the nuclear magnetic resonance (NMR) method for investigating RNA.<\/p>\n\n\n\n<p>In the same way that a single piece of a\npuzzle fits into the whole, the molecule hypoxanthine binds to a ribonucleic\nacid (RNA) chain, which then changes its three-dimensional shape within a\nsecond and in so doing triggers new processes in the cell. Thanks to an\nimproved method, researchers are now able to follow almost inconceivably tiny\nstructural changes in cells as they progress \u2013 both in terms of time as well as\nspace. The research group led by Professor Harald Schwalbe from the Center for\nBiomolecular Magnetic Resonance (BMRZ) at Goethe University has succeeded,\ntogether with researchers from Israel, in accelerating a hundred thousand-fold the\nnuclear magnetic resonance (NMR) method for investigating RNA.<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"alignright size-large is-resized\"><img fetchpriority=\"high\" decoding=\"async\" src=\"https:\/\/aktuelles.uni-frankfurt.de\/wp-content\/uploads\/2020\/01\/beitragsbild_schwalbe.jpg\" alt=\"\" class=\"wp-image-41759\" width=\"407\" height=\"282\" srcset=\"https:\/\/aktuelles.uni-frankfurt.de\/wp-content\/uploads\/2020\/01\/beitragsbild_schwalbe.jpg 650w, https:\/\/aktuelles.uni-frankfurt.de\/wp-content\/uploads\/2020\/01\/beitragsbild_schwalbe-300x208.jpg 300w\" sizes=\"(max-width: 407px) 100vw, 407px\" \/><figcaption><em>Frankfurt researchers followed the movements of this tiny molecule \u2013 just two-thousandths of the thickness of a piece of paper. The RNA aptamer changes its structure when it binds hypoxanthine.  The green nucleobases change shape particularly quickly, the ones coloured blue more slowly. The grey regions do not change. <\/em><\/figcaption><\/figure><\/div>\n\n\n\n<p>\u201cThis allows us for the first time to\nfollow the dynamics of structural changes in RNA at the same speed as they\noccur in the cell,\u201d says Schwalbe, describing this scientific breakthrough, and\nstresses: \u201cThe team headed by Lucio Frydman from the Weizmann Institute in\nIsrael made an important contribution here.\u201d<\/p>\n\n\n\n<p>The new types of NMR experiments use water\nmolecules whose atoms can be followed in a magnetic field. Schwalbe and his\nteam produce hyperpolarized water. To do so, they add a compound to the water\nwhich has permanently unpaired electron radicals. The electrons can be aligned\nin the magnetic field through excitation with a microwave at -271\u00b0C. This\nunnatural alignment produces a polarization which is transferred at +36\u00b0C to\nthe polarization of the hydrogen atoms used in the NMR. Water molecules\npolarized in this way are heated in a few milliseconds and transfered, together\nwith hypoxanthine, to the RNA chain. The new approach can in general be applied\nto observe fast chemical reactions and refolding changes in biomolecules at\natomic level.<\/p>\n\n\n\n<p>In particular the imino groups in RNA can\nbe closely analyzed using this method. In this way, the researchers were able\nto measure structural changes in RNA very accurately. They followed a small\npiece of RNA from <em>Bacillus subtilis<\/em>, which changes its structure during hypoxanthine\nbinding. This structural change is part of the regulation of the transcription\nprocess, in which RNA is being made from DNA. Such small changes at molecular\nlevel steer a large number of processes not only in bacteria but also in\nmulticellular organisms and even humans.<\/p>\n\n\n\n<p>This improved method will in future make\nit possible to follow RNA refolding in real time \u2013 even if it needs less than a\nsecond. This is possible under physiological conditions, that is, in a liquid\nenvironment and with a natural molecule concentration at temperatures around 36\n\u00b0C. \u201cThe next step will now be not only to study single RNAs but hundreds of\nthem, in order to identify the biologically important differences in their refolding\nrates,\u201d says Boris F\u00fcrtig from Schwalbe\u2019s research group.<\/p>\n\n\n\n<p class=\"has-background has-very-light-gray-background-color\"><strong>Publication:<\/strong> Mihajlo Novakovic, Gregory L.
