Graphcis: Michael Schmidt, German Red Cross Blood Donor Service, Frankfurt Institute<\/figcaption><\/figure><\/div>\n\n\n\n<p>Researchers\nat the German Red Cross Blood Donor Service in Frankfurt headed by Professor\nErhard Seifried, and the Institute for Medical Virology at the University\nHospital Frankfurt at Goethe University headed by Professor Sandra Ciesek\nsucceeded in developing a procedure that makes it possible to immediately and\ndramatically increase worldwide testing capacities for detecting SARS-CoV-2.\n\u201cThis makes it possible for the implementation of expanded testing in larger\npopulation groups as demanded by all scientists and also politicians to be\ncarried out sooner than previously thought, even in view of limited test kit\nresources,\u201d says Seifried. The background of this news are laboratory\ninvestigations in which swab samples from mucous membranes of the throat or\nnose are combined using specified procedures in a buffer solution, and\nsubsequently tested using what is known as the PCR procedure (polymerase chain\nreaction procedure, direct genome detection of SARS CoV-2). In the case of a\nnegative result, all included samples have a reliable negative result. The pool\ntesting has no influence on the detection limit. In the case of a positive\nmini-pool result, individual testing is carried out in previously reserved\nsamples. The positive sample can then be identified within 4 hours.<\/p>\n\n\n\n<p>Figure 1\nshows the new mini-pool method. The swab is first placed in a reserve test tube\nand subsequently in a pool container. Since this pool method does not increase\nthe volume in the pool container, no dilution and therefore no decrease in\nsensitivity is observed. Using independent ring trial samples (pre-test of the\nplanned ring trial), that were provided by a ring trial organisation (INSTAND) authorised\nby the by the German Medical Association, it was able to be shown that the new\nmini-pool method can obtain laboratory results of equal quality as those obtained\nwith individual testing. The new method was additionally investigated in a small\nfield study on 50 unselected patient samples. The patient samples were pooled\nin 10 mini-pools of 5 samples each, and also tested individually in parallel.\nOf the 50 patient samples, 5 samples were SARS CoV-2 positive. These samples\nwere distributed across 4 pools. All four mini-pools resulted in a positive PCR\nresult. Mini-pools containing only samples from patients without SARS CoV-2\nalways resulted in a negative result.<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-large\"><img decoding=\"async\" width=\"650\" height=\"245\" src=\"https:\/\/aktuelles.uni-frankfurt.de\/wp-content\/uploads\/2020\/03\/mini-pool-methode_tabelle.png\" alt=\"\" class=\"wp-image-43219\" srcset=\"https:\/\/aktuelles.uni-frankfurt.de\/wp-content\/uploads\/2020\/03\/mini-pool-methode_tabelle.png 650w, https:\/\/aktuelles.uni-frankfurt.de\/wp-content\/uploads\/2020\/03\/mini-pool-methode_tabelle-300x113.png 300w\" sizes=\"(max-width: 650px) 100vw, 650px\" \/><\/figure><\/div>\n\n\n\n<p>\u201cOn the\nbasis of these laboratory results, in particular large cohorts and testing on\nasymptomatic individuals can be carried out, allowing a massive savings of test\nkits in particular,\u201d says Ciesek. The research group is currently analysing\noptions for further enlarging the pools. \u201cThe results give cause for optimism\nthat when globally established, the procedure will quickly enable better\ninformation about the number of people actually infected,\u201d explains Professor\nSchmidt from the Blood Donor Service. This allows the current number of\napproximately 40,000 tests per day in all of Germany to be immediately increased\nto 200,000 to 400,000 tests without reducing the high quality of the diagnosis.\nTo continue efficiently implementing the strategy of early detection and\nisolation, an extension of screening tests is essential, especially for system-relevant\nprofessions such as doctors, nurses, police, firefighters, nursing homes,\nadministration and the food industry. The rights to the invention, for which a\npatent is pending in Europe and the USA, is held jointly by Goethe University\nand the German Red Cross. The technology can be made available immediately to\nother interested institutions through Goethe University\u2019s knowledge transfer\ncompany Innovectis.<\/p>\n\n\n\n<p>Goethe\nUniversity President Professor Birgitta Wolff describes the new analysis method\nas a \u201cmilestone\u201d. \u201cThe more people who can be reliably tested for SARS-CoV-2,\nthe faster the pandemic can be curbed.\u201d<\/p>\n\n\n\n<p>\u201cWith the new method, Germany can take the global fight against SARS-CoV-2 to a new a new level,\u201d observes Seifried with optimism.
