Thanks to this \u201cbait\u201d, the desired protein is available much\nfaster for further examination in the electron microscope. The research team\nhas christened this innovative layer of ultrathin molecular carbon the \u201csmart\nnanosheet\u201d. With the help of this new development, diseases and their treatment\nwith drugs can be better understood, for example.<\/p>\n\n\n\n<p>\u201cWith our process, new types of proteins\ncan be isolated from mixtures and characterized within a week,\u201d explains Daniel\nRhinow from the Max Planck Institute of Biophysics in Frankfurt. \u201cTo date, just\nthe isolation of the proteins was often part of a doctorate lasting several\nyears.\u201d Together with Andreas Terfort (Goethe University) and Andrey Turchanin\n(Friedrich Schiller University Jena), the idea evolved a few years ago of\nfishing the desired proteins directly out of mixtures by equipping a nanosheet\nwith recognition sites onto which the target protein bonds. The researchers\nhave now succeeded in making proteins directly available for examination using\nelectron cryo-microscopy through a \u201csmart nanosheet\u201d.<\/p>\n\n\n\n<p>Electron cryo-microscopy is based on the\nshock-freezing of a sample at temperatures under -150 \u00b0C. In this process, the\nprotein maintains its structure, no interfering fixing and coloring agents are needed,\nand the electrons can easily irradiate the frozen object. The result is high-resolution,\nthree-dimensional images of the tiniest structures \u2013 for example of viruses and\nDNA, almost down to the scale of a hydrogen atom.<\/p>\n\n\n\n<p>In preparation, the proteins are\nshock-frozen in an extremely thin layer of water on a minute metal grid. Previously,\nsamples had to be cleaned in a complex procedure \u2013 often involving an extensive\nloss of material \u2013 prior to their examination in an electron microscope. &nbsp;The electron microscopy procedure is only successful\nif just one type of protein is bound in the water layer.<\/p>\n\n\n\n<p>The research group led by Turchanin is now\nusing nanosheets that are merely one nanometer thick and composed of a cross-linked\nmolecular self-assembled monolayer. Terfort\u2019s group coats this nanosheet with a\ngelling agent as the basis for the thin film of water needed for freezing. The\nresearchers then attach recognition sites (a special nitrilotriacetic acid group\nwith nickel ions) to it. The team led by Rhinow uses the \u201csmart nanosheets\u201d treated\nin this way to fish proteins out of a mixture. These were marked beforehand\nwith a histidine chain with which they bond to the recognition sites; all other\ninterfering particles can be rinsed off. The nanosheet with the bound protein\ncan then be examined directly with the electron microscope.<\/p>\n\n\n\n<p>\u201cOur smart nanosheets are particularly\nefficient because the hydrogel layer stabilizes the thin film of water required\nand at the same time suppresses the non-specific binding of interfering\nparticles,\u201d explains Julian Scherr of Goethe University. \u201cIn this way,\nmolecular structural biology can now examine protein structures and functions\nmuch faster.\u201d The knowledge gained from this can be used, for example, to\nbetter understand diseases and their treatment with drugs.<\/p>\n\n\n\n<p>The team has patented the new nanosheets and additionally already found a manufacturer who will bring this useful tool onto the market.<\/p>\n\n\n\n<p class=\"has-background has-very-light-gray-background-color\"><strong>Publikation:<\/strong> Smart Molecular Nanosheets for Advanced Preparation of Biological Samples in Electron Cryo-Microscopy, ACS Nano 2020, <a href=\"https:\/\/doi.org\/10.1021\/acsnano.0c03052\" target=\"_blank\" rel=\"noreferrer noopener\" aria-label=\" (\u00f6ffnet in einem neuem Tab)\">https:\/\/doi.org\/10.1021\/acsnano.0c03052<\/a><\/p>","protected":false},"excerpt":{"rendered":"<p>An interdisciplinary team from Frankfurt and Jena has developed a kind of bait with which to fish protein complexes out of mixtures.
